The Properties and Specifications of Fluorescein Sodium or Fluorescein Disodium:
Fluorescein Sodium BP Ph Eur Grade Specifications:
Soluble Fluorescein
C20H10Na2O5 --- 376.3 CAS 518-47-8
Action and use: Detection of corneal lesions, retinal angiography and pancreatic function testing.CHARACTERS
Appearance: Orange-red, fine powder, hygroscopic.
Solubility: Freely soluble in water, soluble in ethanol (96 per cent), practically insoluble in hexane and in methylene chloride.IDENTIFICATION
First identification: B, D
Second identification: A, C, D
A. Dilute 0.1 mL of solution S (see Tests) to 10 mL with water. The solution shows yellowish-green fluorescence. The fluorescence disappears on addition of 0.1 mL of dilute hydrochloric acid and reappears on addition of 0.2 mL of dilute sodium hydroxide solution.
B. Infrared absorption spectrophotometry.
C. The absorption by a piece of filter paper of 0.05 mL of the solution prepared for identification A (before the addition of dilute hydrochloric acid) colours the paper yellow. On exposing the moist paper to bromine vapour for 1 min and then to ammonia vapour, the colour becomes deep pink.
D. Ignite 0.1 g in a porcelain crucible. Dissolve the residue in 5 mL of water and filter. 2 mL of the filtrate gives reaction (a) of sodium.TESTS
Solution S: Dissolve 1.0 g in carbon dioxide-free water prepared from distilled water and dilute to 50 mL with the same solvent.
Appearance of solution: Solution S is clear and orange-yellow with yellowish-green fluorescence.
pH:7.0 to 9.0 for solution S.
Related substances: To pass the test by liquid chromatography.
Chlorides: Maximum 0.25 per cent.
Sulfates: Maximum 1.0 per cent.
Zinc: Dilute 5 mL of solution S to 10 mL with water. Add 2 mL of hydrochloric acid, filter and add 0.1 mL of potassium ferrocyanide solution. No turbidity or precipitate is formed immediately.
Loss on drying: Maximum 10.0 per cent, determined on 1.000 g by drying in an oven at 105C.

Fluorescein Sodium contains NLT 90.0% and NMT 102.0% of fluorescein sodium (C20H10Na2O5), calculated on the anhydrous basis.IDENTIFICATION
A. A solution is strongly fluorescent, even in extreme dilution. The fluorescence disappears when the solution is made acid and reappears when the solution is again made alkaline.
B. Identification Tests—General, Sodium: The residue remaining after incineration meets the requirements.
C. The retention time of the major peak of the Sample solution corresponds to that of the Standard solution, as obtained in the Assay.Zinc:
Sample solution: 10 mg/mL of Fluorescein Sodium in a saturated solution of sodium chloride.
Analysis: To 10 mL of Sample solution add 2 mL of 3 N hydrochloric acid. Shake well, filter, and add 1 mL of potassium ferrocyanide to the filtrate.
Acceptance criteria: No turbidity is produced.
Acriflavine:
Sample solution: 2 mg/mL of Fluorescein Sodium in water. Swirl the mixture, and filter.
Analysis: To the Sample solution add a few drops of 100-mg/mL sodium salicylate solution.
Acceptance criteria: No precipitate is formed.
Organic Impurities: To pass the test by chromatography.
Water Determination: NMT 17.0%
Packaging and Storage: Preserve in tight containers.
